Ok, not a mason at all but I have been in construction all my life. First (in most cases) you will have to go with the plumb-ness of the building. You have no choice because someone before you determined that anyway. Run a story pole from the bottom to the top to keep it inline with existing conditions.

If you are building a true block building then you just need to make sure the corners are plumb. Run your plumb corners first and then fill in between them. You will have some leeway with your joints if you need to adjust.
