You have to give the penicillin time to work - one shot is not sufficient. That's why doctors tell you to finish ALL your antibiotics, not stop when you feel better. You can worry about her rumen function later. I never give less than a 5-day course of antibiotics. Probiotics will do the most good once she's off the antibiotics, but it won't hurt to give them now.

Banamine won't hurt her either. I give 1/2 cc per 50 lbs, usually every other day unless it's something really serious, in which case they get it every day for 2-3 days.

Try to get the Thiamine and give that to her until she is 100% - the vit b complex doesn't have near the amounts needed - though it can work and does to some good - also, ask the vet to lt you buy a bottle to keep on hand.
